Do glucosamine and chondroitin help knee pain? What the trials show

Several meta-analyses find glucosamine or chondroitin eases knee pain a little. The largest trials found little benefit over placebo.

Published October 2026

Short answer

Possible small pain relief for knee osteoarthritis, but the evidence is limited and mixed; the largest trials found little beyond placebo. Evidence grade C (Limited). Several meta-analyses of placebo-controlled trials; downgraded for inconsistency (the largest trial and a 2010 network meta-analysis found no clinically meaningful effect) and for low-quality, partly industry-funded trials.

Key takeaways

  • Several meta-analyses of placebo-controlled trials found glucosamine or chondroitin reduced knee pain by a small amount [1][2].
  • The largest trial, GAIT with 1,583 people, found neither supplement significantly reduced knee pain more than placebo over 24 weeks [3].
  • A 2010 network meta-analysis found pain differences below the threshold for a meaningful change, and smaller effects in independent trials than in industry-funded ones [4].

The short version

Glucosamine and chondroitin may ease knee pain a little. Several meta-analyses of placebo-controlled trials found small pain relief [1][2], but the largest trial and a major network review found no clear benefit over placebo [3][4].

What the studies show

In a 2018 meta-analysis, glucosamine reduced knee pain on a visual scale by about 7 mm more than placebo, and chondroitin by about 8 mm; the combination of the two did not show this [1]. A 2024 meta-analysis of 25 trials found chondroitin reduced pain and improved physical function, while glucosamine slowed joint space narrowing seen on X-ray [5]. A Cochrane review of chondroitin found pain scores about 8 points better (on a 0 to 100 scale) than placebo in trials lasting up to six months, though the authors rated the evidence as low quality [6].

The largest trial, GAIT, randomised 1,583 people with knee osteoarthritis to glucosamine, chondroitin, both, celecoxib or placebo for 24 weeks. None of the supplements was significantly better than placebo at producing a 20% drop in pain [3]. In people with moderate to severe pain at the start, the combination did better than placebo (79.2% vs 54.3% responding), but this was an exploratory analysis [3]. Over two years, no treatment reached a clinically important difference in pain or function compared with placebo [7].

The nuances

Who funded a trial seems to matter. In a 2010 network meta-analysis of 10 trials and 3,803 people, independent trials showed smaller effects than industry-funded ones [4]. A 2021 review of 28 trials lasting at least a year found statistically significant but clinically questionable long-term benefits for glucosamine and chondroitin [8].

The 2025 review pooled trials of several products, including plant extract mixes, so its result does not apply to glucosamine or chondroitin alone [2]. In a two-year trial of 605 people, the glucosamine-chondroitin combination slowed joint space narrowing by 0.10 mm more than placebo, a small difference [9].

What this means for you

If you have knee osteoarthritis and are thinking about these supplements, the best reading is a possible small amount of pain relief, with most of the largest studies finding none. Talk with your doctor first, especially if you take medicines for blood sugar, because glucose tolerance changed in one trial of a glucosamine and chondroitin product [10]. Across 69 trials of supplements for osteoarthritis, safety outcomes did not differ from placebo, except for diacerein [11].
